Hastalığın tedavisi; hastanın yaşı, kanserin evresi ve genel sağlık durumu gibi faktörlere bağlı olarak belirlenmektedir. Güncel tedavi seçenekleri arasında cerrahi müdahale, radyasyon tedavisi, hormon tedavisi, kemoterapi ve immünoterapi yer almaktadır. Kanser immünoterapisi, immün sistemin kanser hücrelerini tanıma ve yok etme kapasitesini artırmayı amaçlayan yenilikçi bir tedavi yöntemidir. Bu sayede, kanser hücrelerine karşı daha güçlü ve daha spesifik bir immün yanıt oluşturulması hedeflenmektedir. Son yıllarda yapılan çalışmalar, mikroRNA’ların (miRNA’lar) kanser gelişimi, progresyonu ve immün yanıtın düzenlenmesinde kritik rol oynadığını ortaya koymuştur. miRNA’lar, özellikle prostat kanserinin tanı ve tedavisi için önemli bir biyobelirteç ve potansiyel bir immünoterapi aracıdır. Bu tez çalışmasında, prostat kanseri tedavisi için potansiyel bir immünoterapi stratejisi olarak, miR-30c transfeksiyonu ile stimüle edilmiş NK-92 (sNK-92) hücrelerinin, PNT1A normal prostat hücresi ve PC3 prostat kanseri hücresi üzerindeki etkisinin araştırılması amaçlanmıştır. Çalışma kapsamında sNK92 hücrelerinin sitotoksik potansiyeli, NK-92 ile karşılaştırmalı olarak analiz edilmiştir. Hedef (PC3/PNT1A) ve Efektör (NK/sNK-92) hücreler 1:8 ve 1:16 oranlarında ko-kültür edilmiş ve WST-1 canlılık testi ile değerlendirilmiştir. WST-1 analizini doğrulamak için İmmün boyama analizi yapılmıştır. Elde edilen bulgular, özellikle 1:16 oranında kullanılan sNK-92 hücrelerinin, PC3 prostat kanser hücreleri üzerinde kontrol grubuna kıyasla anlamlı derecede yüksek bir sitotoksik etki yarattığını göstermiştir. Aynı konsantrasyondaki sNK-92 hücreleri, PNT1A normal prostat hücreleri üzerinde herhangi bir sitotoksik etki oluşturmamıştır. Sitotoksik etkinin doğrulanması amacıyla gerçekleştirilen immün boyama analizleri, sNK-92 hücreleri ile ko-kültüre edilen PC3 hücrelerinde immün aktivasyon ve hücre ölümü ile ilişkili sinyallerin arttığını ortaya koymuştur. Normal prostat hücrelerinde ise anlamlı bir immün yanıt veya sitotoksisite gözlenmemiştir.Prostate cancer is a common type of cancer observed in elderly men. The treatment of the disease is determined based on factors such as the patient’s age, the stage of the cancer, and overall health status. Current treatment options include surgical intervention, radiation therapy, hormone therapy, chemotherapy, and immunotherapy. Cancer immunotherapy is an innovative treatment approach that aims to enhance the ability of the immune system to recognize and eliminate cancer cells. In this way, the generation of a stronger and more specific immune response against cancer cells is targeted. Recent studies have demonstrated that microRNAs (miRNAs) play critical roles in cancer development, progression, and the regulation of immune responses. miRNAs are considered important biomarkers and potential immunotherapeutic tools, particularly for the diagnosis and treatment of prostate cancer. In this thesis study, as a potential immunotherapy strategy for prostate cancer treatment, the effects of NK-92 (sNK-92) cells with enhanced cytotoxic activity through miR-30c transfection on PNT1A normal prostate cells and PC3 prostate cancer cells were investigated. Within the scope of the study, the cytotoxic potential of sNK-92 cells was analyzed comparatively with NK-92 cells. Target (PC3/PNT1A) and effector (NK/sNK-92) cells were co-cultured at ratios of 1:8 and 1:16, and cell viability was evaluated using the WST-1 assay. Immunostaining analysis was performed to validate the WST-1 analysis. The obtained findings demonstrated that sNK-92 cells used particularly at a 1:16 ratio induced a significantly higher cytotoxic effect on PC3 prostate cancer cells compared to the control group. sNK-92 cells at the equivalent concentration did not induce any cytotoxicity in normal PNT1A prostate cells. Immunostaining analyses performed to validate the cytotoxic effect revealed increased immune activation and signals associated with cell death in PC3 cells co-cultured with sNK-92 cells.
