Öğe Contribution to the cytotaxonomic knowledge of 10 species of Klasea Cass. (Asteraceae) from Turkey(Firenze Univ Press, 2015) Martin, Esra; Dogan, Bekir; Duran, Ahmet; Coskun, Fatih; Altinordu, FahimIn the present study, karyotype analyses of 10 taxa belongs to the genus Klasea Cass. (Asteraceae) were conducted. Somatic chromosome numbers are not a distinctive character of classification of Klasea taxa. All of the taxa were counted as 2n = 30 chromosomes, except for Klasea radiata subsp. biebersteiniana (2n = 60). Although all the taxa have a chromosome number of 2n = 30, they could be differentiated by their karyotype formula and quantitative parameters of the karyotype. Karyotype analysis was performed by means of an Image Analysis System and karyotype asymmetry indices were measured. According to these parameters, karyological relationships among studied taxa were inferred.Öğe Determination of Antioxidant, Anticholinesterase, Tyrosinase Inhibitory Activities and Fatty Acid Profiles of 10 Anatolian Klasea Cass. Species(Acg Publications, 2016) Tel, Gulsen; Dogan, Bekir; Erol, Ebru; Ozturk, Mehmet; Nadeem, Said; Ullah, Zain; Duru, Mehmet EminIn search of new natural fatty acid sources, extract of 10 different Turkish Klasea species were studies. Fatty acids of Klasea species were studied by GC and GC-MSD. Oleic acid (4.8-45.8%), palmitic acid (15.6-51.8%), linoleic acid (0.3-45.5%), palmitoleic acid (0.8-28.4%) and linolenic acid (15.6-34.6%) were the main fatty acids elucidated. All extracts were also subjected to acetylcholinesterase, butyrylcholinesterase, tyrosinase, beta-carotene-linoleic acid, DPPH center dot scavenging, CUPRAC and ferrous ion-chelating ability activities. Total flavonoid and phenolic contents were determined as quercetin and pyrocatechol equivalents. All extracts showed significant antioxidant activity in all tests, except hexane extracts of K. serratuloides and K. cerinthifolia that showed weak inhibition against BChE and AChE. The hexane extract of K. coriaceae and methanol extract of K. serratuloides exhibited notable tyrosinase inhibitory activity.Öğe Fatty Acid Composition, Antioxidant, Anticholinesterase and Tyrosinase Inhibitory Activities of Four Serratula Species from Anatolia(Acg Publications, 2013) Tel, Gulsen; Ozturk, Mehmet; Duru, Mehmet Emin; Dogan, Bekir; Harmandar, MansurSerratula L. (Astareceae) rich in ecdysteroid, phytoecdysteroids and flavonoids some have various biological activities including antibacterial and antitumor. The fatty acid profiles of four Serratula species were investigated by using GC and GC-MS techniques. Palmitic, oleic, linoleic and linolenic acids were found to be the main fatty acids. The unsaturation percentage was between 27.24-50.47%. The antioxidant activity of the extracts was determined by using four complementary tests; namely, beta-carotene-linoleic acid, DPPH center dot scavenging, CUPRAC and ferrous-ions chelating assays. The methanol extract of S. lasiocephala showed the highest activity in beta-carotene-linoleic acid, DPPH center dot scavenging and CUPRAC assays, while the hexane extract of S. radiata exhibited the best metal chelating activity. In addition, total phenolic and total flavonoid contents in the extracts were determined as pyrocatechol and quercetin equivalents, respectively. The in vitro anticholinesterase activity of extracts were tested against acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) which are the key enzymes taking place in pathogenesis of Alzheimer's disease. Besides, the extracts were tested in vitro against tyrosinase enzyme which is associated with melanin hyperpigmentation. Except the hexane extracts of S. radiata and S. lasiocephala, the extracts showed moderate inhibition against AChE and BChE, while the only hexane extract of S. erucifolia and methanol extract of S. radiata exhibited tyrosinase inhibitory activity.Öğe Jurinea kemahensis (Asteraceae), a new species from East Anatolia, Turkey(Finnish Zoological Botanical Publishing Board, 2014) Dogan, Bekir; Kandemir, Ali; Osma, Etem; Duran, AhmetA new species Jurinea kemahensis B. Dogan, Kandemir & A. Duran (Asteraceae) is described from East Anatolia, Turkey. It grows in calcareous crevices in the Kemah district (Erzincan province) in the East Anatolia. It is morphologically fairly similar to J. tortumensis, and its diagnostic morphological characters are discussed. The ecology and distribution of J. kemahensis are also presented. Greuter & Wagenitz subsp. karamanica B. Dogan & A. Duran (Asteraceae), is described and illustrated from Anatolia, Turkey. The subspecies grows on stony places in the Ayranci and Ermenek district (C4 Karaman Province), Eregli district (C5 Konya province) and Ulukisla (C5 Nigde province) in the central and south Anatolia transition zone. It is morphologically similar to K. serratuloides subsp. serratuloides. Diagnostic morphological characters from closely similar taxa are discussed and arranged in a key. Ecology and conservation status of the subspecies is also presented. In addition, the geographical distribution of the new subspecies and other related species is mapped. Br. (Brassicaceae) is taxonomically problematic and some of its species have recently been transferred to the genera Strigosella Boiss. and Zuvanda Dvorak. In this study, pollen morphology of some species of Malcolmia, Strigosella and Zuvanda, previously placed in illalcolmia genus, were examined under scanning electron microscope (SEM), in order to determine the significance of pollen features as taxonomic characters. The results reveal rather uniform morphological features, however fine details are characteristic to distinguishing pollen grains in the species of the genus. The pollen grains in three genera are tricolpate and the surface sculpturing type is reticulate. The basic shape of the pollen grains in species studied is euprolate, subprolate-euprolate and euprolate-perprolate. While pollen grains of S. africana is the smallest in all examined species, M. chic is the.biggest. However, three genera can be easily separated by the length of the polar and equatorial axes themselves.Öğe Psephellus vanensis (Asteraceae), a new species from east Turkey(Pensoft Publishers, 2015) Dogan, Bekir; Behcet, Lutfi; Duran, Ahmet; Avlamaz, DavutA new species, Psephellus vanensis A.Duran, Behcet & B.Dogan (Asteraceae) from Anatolia, Turkey, is described and illustrated. The species grows on the serpentine stony field of the village of caldiran in the district of Baskale (Van province) in eastern Anatolia. It is morphologically similar to Psephellus pyrrhoblepharus (Boiss.) Wagenitz. Diagnostic characters are discussed, and a key to the most similar species is provided. Ecology, conservation status and notes on biogeography of the species are also presented. In addition, the geographical distribution of the new species and other related species in Turkey is mapped.Öğe Study of phylogenetic relationship of Turkish species of Klasea (Asteraceae) based on ISSR amplification(Pensoft Publishers, 2015) Dogan, Bekir; Duran, Ahmet; Seker, Meryem; Cetin, Ozlem; Martin, EsraKlasea is a taxonomically complex genus in which there are many problems, mostly with Klasea kotschyi and K. hakkiarica. It is challenging to differentiate the genera based on morphological characters alone. Revision studies performed on the basis of molecular data obtained from studies conducted in recent years have made the phylogenetic relationships and systematic positions of the taxa more apparent and reliable. In this study, Klasea, Serratula, Jurinea and Centaurea species native to Turkey, were collected from different localities of Anatolia and DNA was isolated from the collected samples. The data were analyzed ordination analyses including UPGMA and PCA using NTSYSpc 2.1. The infrageneric and intergeneric phylogenetic relationships between Klasea and other related genera were also characterized. The Klasea species were grouped into three clusters. It was determined that taxa Klasea kotschyi and K. hakkiarica are separate but closely related. Moreover, it was observed that the Klasea lasiocephala a separate group within the genera. Clearly the genera Klasea, Serratula, Jurinea and Centaurea are phylogenetically differentiated on the dendogram.Öğe Study of phylogenetic relationship of Turkish species of Matthiola (Brassicaceae) based on ISSR amplification(Tubitak Scientific & Technological Research Council Turkey, 2016) Dogan, Bekir; Celik, Mustafa; Unal, Murat; Sefali, Abdurrahman; Martin, Esra; Kaya, AylaMatthiola W.T. Aiton is a taxonomically complex genus in which there are many problems, mostly with Matthiola longipetala and M. odoratissima. Matthiola species native to Turkey were collected from various locations in Anatolia, and their DNA was isolated. Revision studies performed on the basis of molecular data obtained from studies conducted in recent years have made the phylogenetic relationships and systematic positions of the taxa more apparent and reliable. Consequently, the remaining taxonomic problems among the species have been resolved through the use of DNA-based molecular analysis methods, which, unlike phenotype studies, are not affected by environmental factors. The inter-simple sequence repeat (ISSR) fingerprinting method was used in the study because its properties were considered to be more reliable and consistent than those of the randomly amplified polymorphic DNA method. DNA fragments were amplified through the use of ISSR primers. The phylogenetic relationships among the taxa were represented on a dendrogram constructed through means of NTSYSpc 2.02 software. The infrageneric and intergeneric phylogenetic relationships between Matthiola and other related genera were also characterized. It was determined that the taxa Matthiola odoratissima and M. ovatifolia are separate but closely related. Moreover, it was observed that the Matthiola longipetala complex forms a separate group within the genera. Clearly, the genera Matthiola, Sterigmostemum, Strigosella, Malcolmia, and Chorispora are phylogenetically differentiated on the dendrogram.
