Öğe Antiviral activity of Ribes uva-crispa L. extracts in vitro(Univ Karachi, 2020) Dogan, Hasan Huseyin; Duman, Rustem; Dinc, MuhittinThere is currently no approved vaccine or a useful antiviral drug against respiratory syncytial virus (RSV) that causes viral infection worldwide. Crude plant extracts can be an important resource for the development of new anti-RSV agents. In this study, cytotoxic and anti-RSV effect of the extracts Ribes uva-crispa, which has been known as gooseberry in Turkey and fruits used in the treatment of the various disorders, were evaluated by colorimetric XTT method. Results were expressed as 50% cytotoxicity (CC50), 50% effective concentration (EC50) and selectivity index (SI: CC50 / EC50). Of the tested extracts, the highest antiviral activity was found to be 96.90 mu g/mL EC50 and 11.70 SI from fruit aqueous extract; it was followed by leaf methanol extract (EC50: 2527.41 mu g/mL, SI: 6.55), leaf aqueous extract (EC50: 1093.37 mu g/mL, SI: 1.40) and fruit methanol extract (EC50: 11262.35 mu g/mL, SI: 0.56), respectively. As a result, we can say that these extracts, especially Ribes uva-crispa fruit aqueous and leaf methanol extracts, are worthy of further studies for the development of new and unique anti-RSV drugs.Öğe CYTOTOXIC AND ANTIVIRAL ACTIVITY OF RIBES UVA CRISPA LINN. EX ROMER AND SCHULTES EXTRACTS(Int Journal Pharmaceutical Sciences & Research, 2018) Duman, Rustem; Dogan, Hasan Huseyin; Dinc, Muhittin; Tuncer, PinarTo find new antiviral agents from natural sources, methanol and aqueous extracts of leaves and fruits of Ribes uva crispa and Ribes multiflorum naturally grown in Turkey were investigated as in vitro to reveal their antiviral activities against Herpes simplex virus type 1. All experiments were performed in 96-well plates and results were measured with XTT-based colorimetric assay. Results demonstrated that all extracts have no cytotoxic effect on vero cells at 10000 mu g/mL, while they also exhibited anti-HSV-1 activity with different percentages of protection (varying between 2.65% - 50.40%) in 10000 mu g/mL which was at the highest concentration in vero cells. 50% effective concentrations (EC50) of the extracts which were determined having percentage of protection against HSV-1 at concentrations lower than 10000 mu g/mL were calculated using GraphPad Prism Version 5.03 statistics program with non-linear regression analysis. These extracts were determined to have EC50 values ranging between 9710 - 70600 mu g/mL and selectivity index (SI) are ranging between 0.14 - 1.03.
