Tabletlerde Parasetamol ve Tramadol Hidrokloridin Ana Bileşen Regresyonu ve Kısmi En Küçük Kare Yöntemleriyle Eş Zamanlı Spektrofotometrik Tayini

Özet
İlaç endüstrisi ürünlerinin çeşitliliği gerek doz gerekse de kullanılacak olan ilaç içeriklerinin
farmasötik ve tıbbi araştırma süreçleri sonrasında ortaya çıkarılan tedavilerin süre ve maliyeti, günümüz
şartlarında çok önemlidir. Özellikle tedavisi güç ve zaman alan hastalıklara karşı üretilen ve kullanılan
kombine ilaç içeriklerinin miktar tayinlerinde, analitik yöntemler önemli araçlardır ve bu yöntemlere
ihtiyaç giderek artmaktadır. Kombine ilaçlar hedefe yönelik terapinin en önemli araçlarındandır ve her
geçen gün daha fazla sayıda hastanın yaşamına katkıda bulunmaya devam etmektedir. Bu ve benzeri tip
müstahsarların içerik ve miktar tayinine dönük kantitatif tayinlerde, en çok kullanılan kemometrik
yöntemlerden ikisi ana bileşen regresyonu (PCR) ve kısmi en küçük kareler (PLS) dir. Bu yöntemler
matematiksel olarak ele alındığında her bir müstahzarın miktar tiyinine dönük aletli analiz
yöntemlerinden elde edilen dataların analizinde, doğru, güvenilir ve hızlı sonuçlar elde etmeye imkan
sağlar.
Bu tez çalışmasında kombine bir ticari tablet formülasyonu olan Zaldiar’ın içeriğini teşkil eden,
tramadol hidroklorür (TRA) ve parasetamolün (PAR) eşzamanlı tayini, miktar tayini, ekstraksiyon veya
ön ayırma gibi işlemler kullanılmadan karışımdan elde edilen UV-spektrofotometrik verilerine ana
bileşen regresyonu ve kısmi en küçük kareler yöntemleri uygulanarak gerçekleştirildi. Analitlerin
spektrofotometrik tayini için, PAR için 6-36 µg/mL ve TRA için 4-10 µg/mL doğrusal konsantrasyon
aralıklarında; PAR ve TRA içeren 16 farklı karışımdan oluşan bir kalibrasyon seti, sırasıyla 24
tam
fraksiyonel tasarım modeli kullanılarak hazırlandı. Örneklerin absorbansları 215-280 nm (∆λ=0.1 nm)
arasında ölçüldü. Kalibrasyon seti için absorbans veri seti 215-280 nm (∆λ=0.1 nm) arasındaki
absorbsiyon spektrumlarından gelen datalar kullanılarak elde edildi, devamında konsantrasyon ve
absorbans setleri kullanılarak PCR ve PLS kalibrasyonu yapıldı.
Deney sonuçlarının kemometrik yöntemlerle karşılaştırılması amacıyla, PAR/TRA ikili
kombinasyonlarını içeren karışımların kantitatif çözünürlüğü için oran spektrumları-birinci türev
spektrofotometrisi geliştirilmiştir. Önerilen kemometrik yöntemler PCR ve PLS ve oran spektrumları birinci türev yaklaşımları validasyon örnekleri incelenerek doğrulandı ve ticari tablet örneklerinin
analizine uygulandı. Analiz sonuçları, önerilen yöntemlerin TRA ve PAR’ın ikili karışımlarını içeren
ticari tablet formülasyonlarının kalite kontrol ve rutin analizlerinde kantitatif analiz için çok uygun
olduğunu göstermiştir.
The diversity of the products of the pharmaceutical industry, both the dose and the drug ingredients to be used, the duration and cost of the treatments revealed after the pharmaceutical and medical research processes are very important in today's conditions. Analytical methods are important tools for the determination of the ingredients of combined drugs produced and used especially against diseases that are difficult to treat and ineffective, and the need for these methods is increasing. Combined drugs are one of the most important tools of targeted therapy and continue to contribute to the lives of more and more patients. The most commonly used chemometric methods for quantification are principal component regression (PCR) and partial least squares (PLS). These methods, when considered mathematically, allow to calculate how much each preparation is absorbed and to obtain fast results. In this thesis, which is combined with a commercial tablet formulation Zaldiar the contents of Tramadol Hydrochloride (TRA) and paracetamol (PAR) simultaneous determination assay, extraction or pre-separation processes such as obtained from the mixture without the use of UV-spectrophotometric methods was performed by applying principal component and partial least squares regression to the data. For the spectrophotometric determination of analytes, a calibration set consisting of 16 different mixtures containing PAR and TRA in linear concentration ranges of 6-36 µg/mL for PAR and 4-10 µg/mL for TRA was prepared as 24 complete fractional designs, respectively. The absorbances of the samples were measured between 215-280 nm (∆λ=0.1 nm). The absorbance data set for the calibration set was obtained between 215-280 nm (∆λ=0.1 nm). PCR and PLS calibration were performed using concentration and absorbance sets. To compare the experimental results with chemometric methods, ratio spectra-first derivative spectrophotometry was developed for the quantitative resolution of binary mixtures containing PAR-TRA combinations. The proposed chemometric methods (PCR and PLS) and ratio spectra-first derivative approaches were validated by examining validation samples and applied to the analysis of commercial tablet samples. The analysis results showed that the proposed methods are very suitable tools for the routine analysis and quantification of TRA and PAR in commercial tablets.
