Özet
Bu tezde, IrO2 nanopartiküller (IrO2NP) ile katkılanmış bakteriyel selüloz (BS) kullanılarak
dopamin tespitinde kullanılmak üzere kâğıt temelli bir optik sensör hazırlanmıştır. Elde edilen kompozit
nanomalzeme (IrO2NP@BS) Fourier Dönüşümlü Kızılötesi Spektroskopisi (FT-IR), Raman Spektroskopisi,
X-ışını Kristalografisi (XRD), Geçirimli Elektron Mikroskobu (TEM), Alan Emisyonlu Taramalı Elektron
Mikroskobu (FE-SEM), Atomik Kuvvet Mikroskobu (AFM) ve UV-Vis-NIR Spektrofotometresi ile
karakterize edilmiştir.
Hazırlanan kompozit nanomalzemedeki IrO2NP için iki farklı sentez yöntemi kullanılmıştır. Uygun
sentez yönteminin belirlenmesiyle beraber liyofilizasyon ve süzme metotları ile iki farklı kâğıt platform
uygulaması gerçekleştirilmiştir. Burada, sentezlanen IrO2NP’ler ile BS katkılanarak kompozit malzeme elde
edilmiştir. Kâğıt ortamında, katekol grubunda yer alan dopaminin IrO2 nanopartikül yüzeyleri üzerindeki
olası bağlanması sonucu renk değişiklikleri araştırılmıştır. Ayrıca, IrO2NP@BS ile elde edilen kâğıt
sensörün askorbik asit, ürik asit, fenilalanin, glikoz, lizin, histidin, sistein ve sistin gibi biyomoleküllere
karşı seçicilik çalışmaları da yapılmıştır. Deneyler neticesinde kâğıt sensörün dopamine karşı seçici olduğu
tespit edilerek hızlı test kiti uygulaması gerçekleştirilmiştir. Sonuç olarak, farklı biyoaktif mololeküllerle
yapılan bu çalışmada, IrO2NP@BS’nin optik özellikleri, taşıma ve uygulama kolaylığı, sunmuş olduğu
imkânlar sayesinde farklı alanlarda dopamin sensör olarak kullanılabilirliğini gösterilmiştir.
In this thesis, a paper-based optical sensor for dopamine detection was obtained by using bacterial cellulose (BS) doped with IrO2 nanoparticles (IrO2NP). The resulting composite nanomaterial (IrO2NP@BS) was characterized by Fourier Transform Infrared Spectroscopy (FT-IR), Raman Spectroscopy, X-ray Crystallography (XRD), Transmission Electron Microscope (TEM), Field Emission Scanning Electron Microscopy (FE-SEM), Atomic Force Microscopy (AFM) and UV-Vis-NIR Spectrophotometer. Two different IrO2NP synthesis methods were used to add the prepared composite nanomaterials. With the determination of the appropriate synthesis method, two different paper platform applications, lyophilization and filtration methods, were also investigated. Here, composite material was obtained by doping IrO2NP and BS matrices, and color changes were investigated as a result of possible binding of dopamine in the catechol group on the IrO2NP surfaces in the paper medium. For this purpose, synthesis and characterization of homogeneous nanoparticles, obtaining selectivity for dopamine among various interacting compounds and biomolecules (ascorbic acid, uric acid, phenylalanine, glucose, lisin, histidine, cysteine and cystine) based on the interaction of nanoparticle surface, are suitable for the detection of dopamine in vital fluids of the detection limit of sensor application. This study includes objectives such as the observation of dopamine sensor in different areas, thanks to the optical properties of IrO2NP@BS, the possibilities it offers thanks to its easy transport and application.
