Demir Oksit Nanoparçacıklarının Biyosentezi, Karakterizasyonu ve İn Vitro Değerlendirilmesi

Özet
Metalik nanoparçacıklar; biyomedikal uygulamalarda; nano taşıyıcı olarak ilaç dağıtımında,
hastalık tedavisinde, tıbbi görüntülemede ve biyoteknoloji çalışmalarında kullanılabilmesi açısından çok
fazla ilgi çekmektedir. Tıp alanında en çok kullanılan metalik nanoparçacıkları arasında demiroksitler
(FeO, Fe2O3, Fe3O4) bulunmaktadır. Son yıllarda, özellikle biyomedikal alanda kullanılacak olan
demiroksit nanoparçacıkların sentezini biyolojik sistemlerle yapmaya yönelik çalışmalar yapılmaktadır.
Bu amaçla algler, enzimler, bitki özütleri kullanılarak toksisitesi düşük çevre dostu malzemeler
kullanılabilmektedir. Biyolojik orijinli materyallerin kullanımı nanoparçacık yüzeyini sarmaları ile çökme
eğilimini engelleyerek ve toksik etkilerini iyileştirerek onları biyouyumlu hale getirmektedir. Bu tezin
temel amacı, Punica Granatum (nar) kabuğu özütü kullanılarak biyomedikal alanda kullanılabilecek çok
küçük boyutlarda ve kararlılıkta demir oksit nanoparçacıklarının sentezi, hazırlanan nanoparçacıkların
karakterizasyonunun yapılması ve kanser tedavisindeki etkinliğinin araştırılmasıdır. Sentezlenen
nanoparçacıkların karakterizasyonu Geçirimli Elektron Mikroskobu (TEM), UV-Vis Spektrofotometresi
(UV-Vis), X-ışını Kırınım Spektroskopisi (XRD), Zeta Potansiyeli Ölçümü, Infrared Spektrofotometresi
(FTIR), Termal Gravimetrik Analiz (TGA), Titreşimli Örnek Manyetometrisi (VSM) yöntemleri ile
yapılmıştır. Punica Granatum özütü ile sentezlenen nanoparçacıklar çok küçük boyutta; 3.76 ± 0,59 nm ve
Fe2O3 kristal yapısında oluşmuştur ve sulu çözeltide haftalarca kararlı kalabilmektedir. Nanoparçacıkların
derişim artışına bağlı olarak antioksidan aktivitesinde artış olduğu ve kontrol olarak kullanılan askorbik
asitin maksimum inhibisyon değerine yaklaştığı görülmüştür ve aynı zamanda nanoparçacıklarının E. coli
bakterilerine karşı antibakteriyel aktivitesi göstermiştir. Nanoparçacıkların in vitro değerlendirilmesinde,
meme kanser; MDA-MB-231 ve meme normal; CRL-4010 hücre hatları kullanılmıştır. Nanoparçacıklar,
CRL-4010 hücrelerine göre, MDA-MB-231 hücrelerinde daha fazla toksik etki göstermiştir ve 2,86 kat
artış ile MDA-MB-231 hücrelerinde seçici apoptoza neden olmuştur. Nanoparçacıkların hücre ile
etkileşiminin hücre içine alım ve hücre çeperine bağlanma yoluyla olduğu, hücre ile muamele edilen
nanoparçacıkların Prusya mavisi ile boyanarak görüntülenmesi ile gösterilmiştir. Sonuçlara göre, yeşil
sentez yoluyla elde edilen nanoparçacıkların, potansiyel bir antikanser, antioksidan ve antibakteriyel ajan
olarak literatüre katkı sağlayacağı düşünülmektedir.
Metallic nanoparticles attract much attention as they can be used as nano-carrier in drug delivery, disease treatment, medical imaging in biomedical applications and in biotechnology studies. There exists iron oxides (FeO, Fe2O3, Fe3O4) nanoparticles among the most used metallic nanoparticles in the medical field. In recent years, studies have been carried out to synthesize magnetic nanoparticles that will be used in especially biomedical field with biological systems. For this purpose, more environmentally friendly materials with low toxicity can be obtained by using algae, enzymes, plant extracts. The use of materials of biological origin makes the nanoparticles surface biocompatible by preventing its collapse behavior and improving toxic effects by coating the surface. The fundamental purpose of the thesis is, the synthesis of iron oxide nanoparticles with an ultra-small size and stability for biomedical fields, characterization of nanoparticles and investigation of the effectiveness in cancer therapy. Characterization of nanoparticles synthesized was performed by Transmission Electron Microscope (TEM), UV-Vis Spectrophotometer (UV-Vis), X-ray Diffraction Spectroscopy (XRD), Zeta Potential Measurement, Infrared Spectrophotometer (FTIR), Thermal Gravimetric Analysis (TGA), Vibrating Sample Magnetometer (VSM). Nanoparticles synthesized with Punica Granatum has the crystal form of Fe2O3, with an ultra small size of 3.76 ± 0,59 nm, and are stable for weeks in aqueous solution. It is seen that antioxidant activity of nanoparticles is increased with the concentration of nanoparticles and approached the max. inhibition value of that of ascorbic acid used as control and nanoparticles also showed antibacterial activity to Escherichia coli bacteria. For in vitro evaluation of nanoparticles; breast cancer; MDA-MB 231 and breast normal; CRL-4010 cell lines were used. Nanoparticles displayed more toxic effect on MDA-MB-231cells in contrast to CRL-4010 cells and gave rise to selective apoptosis with a 2.86 fold increase on MDA-MB-231 cells. It is showed that the interaction of nanoparticles with cells is occured by cellular uptake of nanoparticles and binding nanoparticles on the cell membrane and it is visualized by Prussian blue staining of cells incubated with nanoparticles. According to the results, it is expected that nanoparticles synthesized by green ways will be able to contribute as a potential anticancer, antioxidant and antibacterial agent to the literature.
