Deneysel depresyon oluşturulmuş sıçanlarda antidepresan tedavinin hipokampus ve hipotalamus endoplazmik retikulum stresi üzerıne etkilerinin araştırılması

Özet
Amaç: Bugüne kadar Major Depresyonun patofizyolojisini anlamaya dönük olarak çok sayıda
çalışma yapılmış olmasına ragmen altta yatan mekanizma tam olarak netlik kazanamamıştır.
Bu çalışmadaki amacımız ER stresinin, depresyon fizyopatolojisindeki rolünü incelemek ve
mevcut tedavilerdeki geç etki ve tedaviye cevapsızlık sorununu aşmak için ER stres üzerine
direkt etkili yeni tedavi stratejilerinin önünü açmaktır.
Yöntem: Çalışmanın ilk aşamasında sıçanlarda zorlu yüzme testi ile deneysel depresyon modeli
oluşturulmuş ve daha sonra 14 gün boyunca 2 farklı dozda sertralin tedavisi uygulanmıştır.
Sıçanlar kontrol grubu, depresyon grubu, depresyon+ 1 mg sertralin ve depresyon+10 mg
sertraline olmak üzere 4 gruba ayrılmıştır. Ikinci aşamada sıçanların beyin dokuları disseke
edilerek hipokampus ve hipotalamusta ER stres moleküllerinin gen ifadesi ölçülmüştür.
Bulgular: ATF4 gen ifadesinin hipokampusta depresyonda arttığı ve tedavi ile azaldığı,
hipokampal GRP78 gen ifadesinin tedaviden sonra azaldığı, hipotalamik ve hipokampal CALR
gen ifadesinin tedavi ile azaldığı, hipotalamus HSP47 gen ifadesinin her iki tedavi grubunda
azaldığı, hipotalamik ve hipokampal XPB1 gen ifadesinin tedavi ile azaldığı, hipokampal XPB1
gen ifadesinin depresyon +10 mg sertralin grubunda, depresyon+1 mg sertralin grubuna göre
daha da azaldığı bulundu.
Sonuç: Bulgular ER stres mekanizmasının depresyonun patofizyolojisinde rol alabileceğini ve
tedavi ile birlikte bu mekanizmanın geri döndürülebileceğini göstermektedir. Çalışma bulguları
insan çalışmaları için cesaretlendirici olmuş ve antidepresan etkiyi anlamak ve hızlandırmak
için yeni projeler için ufuk açıcı olmuştur. Teyit edilebilmesi ve bu değişimin nasıl olduğunun
gösterilebilmesi için yeni çalışmalara ihtiyaç vardır.
Objective: Until now, the underlying mechanism for understanding the pathophysiology of Major Depression has not been fully clarified. Our aim in this study is to examine the role of ER stress in the physiopathology of depression and to pave the way for new treatment strategies on ER stress in order to overcome the effect of current treatments and non-response to treatment. Method: In the first stage of the study, an experimental depression model was created with the forced swimming test in rats, and then 2 different doses of sertraline treatment were administered for 14 days. The rats were divided into 4 groups as control group, depression group, depression + 1 mg sertraline and depression + 10 mg sertraline. In the second step, the brain tissues of rats were dissected and the gene expression of ER stress molecules in the hippocampus and hypothalamus was measured. Results: It was found that ATF4 gene expression increased in depression in the hippocampus and decreased with treatment, hippocampal GRP78 gene expression decreased after treatment, and hypothalamic and hippocampal CALR gene expression decreased with treatment. Hypothalamus HSP47 gene expression decreased in both treatment groups, hypothalamic and hippocampal XPB1 gene expression decreased with treatment, and hippocampal XPB1 gene expression was further decreased in the depression + 10 mg sertraline group compared to the depression + 1 mg sertraline group. Conclusion: The findings show that the ER stress mechanism may play a role in the pathophysiology of depression and this mechanism can be reversed with treatment. The study findings have been encouraging for human studies and stimulating new projects to understand and accelerate the antidepressant effect. New studies are needed to confirm and show how this change happened.
