Özet
Bu tez çalışması tuz stresi altında mısır (Zea mays L.) köklerinde biyouyarıcı (fosfit) uygulanarak ozmotik düzenleme ve antioksidan kapasite üzerindeki etkilerini araştırmak amacıyla yapılmıştır. Tez çalışmamızda stres faktörü olarak tuz stresi (100 mM) seçilmiştir. Stresle eş zamanlı olarak biouyarıcı olarak sınıflandırılan fosfit (PHI, 1 ve 3 g L-1) uygulanmıştır. Mısır tohumları (DKC 6664), hidroponik ortam ve hoagland besin çözeltisinde, iklimlendirme odasında büyütülmüş olup 21 gün sonra kök hasatı yapılmıştır. Hasat edilen mısır köklerinde su içeriği oranı (RWC), prolin miktarı, protein miktarı, ROS (H2O2 ), lipid peroksidasyonu (TBARS) ve antioksidan enzim /izozim analizleri yapılmıştır. Yapılan analizler sonucunda tuz stresine maruz kalan mısır köklerinde RWC ve protein değerlerinde azalmalar saptanmıştır. Tuz stresi köklerde süperoksit dizmutaz (SOD), katalaz (CAT) ve peroksidaz (POX) enzim aktivitelerini artırmıştır. Bu enzimlerde artışlara rağmen köklerdeki hidrojen peroksit (H2O2) süpürülememiştir. Devamında TBARS düzeylerinde de artışlar izlenmiştir. Dışarıdan uygulanan fosfit, tuz stresine karşı RWC, prolin ve protein miktarlarında artışlara neden olmuştur. Strese maruz kalan mısır köklerine eş zamanlı olarak fosfit eklendiğinde SOD, CAT ve POX enzim aktivitelerinde artışları sağlamıştır. Bunun sonucunda H2O2 etkili olarak ortadan kaldırılmış ve H2O2 ve TBARS miktarlarında azalmalar ölçülmüştür. Tüm bu sonuçlar değerlendirildiğinde tuz stresine karşı fosfit uygulamalarının su içeriği, ozmoregülasyon ve antioksidan savunma sistemi üzerine olumlu etkileri olduğunu söyleyebiliriz.
This thesis study was carried out to investigate the effects on osmotic regulation and antioxidant capacity by applying biostimulant (phosphite) to maize (Zea mays L.) roots under salt stress. In our thesis study, salt stress (100 mM) was chosen as the stress factor. Phosphite (PHI, 1 and 3 g L-1), classified as a biostimulant, was applied concurrently with stress. Maize seeds (DKC 6664) were grown in hydroponic medium and hoagland nutrient solution in a conditioning chamber and root harvested after 21 days. The water content ratio (RWC), proline amount, protein amount, ROS (H2O2), lipid peroxidation (TBARS) and antioxidant enzyme / isozyme analyzes were performed in harvested maize roots. As a result of the analysis, decreases in RWC and protein values were determined in maize roots exposed to salt stress. Salt stress increased superoxide dismutase (SOD), catalase (CAT) and peroxidase (POX) enzyme activities in roots. Despite the increases in these enzymes, hydrogen peroxide (H2O2) in the roots could not be scavenged. Subsequently, increases in TBARS levels were also observed. Externally applied phosphite caused increases in RWC, proline and protein amounts against salt stress. When phosphite was added to the stressed maize roots simultaneously, SOD, CAT and POX enzyme activities were increased. As a result, H2O2 was effectively removed and reductions in the amounts of H2O2 and TBARS were measured. When all these results are evaluated, we can say that phosphite applications against salt stress have positive effects on water content, osmoregulation and antioxidant defense system.
