Özet
Nanoparçacıklar, farklı içerik, boyut ve şekillerde kendilerine has kimyasal reaktivite ve
antimikrobiyal, antienflamatuvar, antiviral, antioksidan, yara iyileştirici ve antikanser gibi farklı
fizikokimyasal özellik ve biyolojik aktivite gösteren malzemelerdir. Gümüş nanoparçacıkların yüksek
antimikrobiyal etkinlik göstermesi, çok spektrumlu bir antibiyotik olması, yüksek derişimlerde bile
toksisitelerinin düşük olması gibi özellikler gümüşü birçok çalışma alanında kullanılabilir kılmaktadır.
Nanoparçacık sentez yöntemlerinden, yeşil sentez kolay ve düşük maliyetli olması bununla birlikte düşük
toksisite göstermesi nedeniyle çevre dostu bir yaklaşım olarak sıklıkla kullanılan bir yöntemdir. Kullanılan
bitki özütleri doğada hâlihazırda bulunan, vücut savunma mekanizmasını yüksek oranda tehdit etmeyen,
biyolojik orijinli ve çok fonksiyonlu nano platformlar hazırlamak için kullanılan materyaller olarak,
özellikle biyoteknoloji ve tıp alanında çok ilgi görmektedir. Bu sebeplerden dolayı gümüş nanoparçacık
sentezinde Cotinus coggygria Scop. (Duman Ağacı) bitkisinin su içindeki ekstraktı kullanılmıştır.
Nanoparçacıkların sentezinde reaksiyon parametreleri optimize edilerek, çok küçük, homojen boyut
dağılımlı ve sulu çözeltisinde kararlı nanoparçacıklar sentezlenebilmiştir, karakterizasyonları için
UV/Görünür Bölge Spetrofotometresi, Elektron Mikroskobu (STEM), Enerji Dağılımlı X-Işını
Spektroskopisi (EDX), Infrared Spektrofotometresi (FTIR), X Işını Kırınımı Difraktometresi (XRD) ve
Zeta Potansiyeli cihazları kullanılmıştır. Sentezlenen gümüş nanoparçacıkların in vitro çalışmaları için üç
meme hücresi hattı (MCF-7, MDA-MB-231 ve CRL 4010) kullanılarak, nanoparçacıkların sitotoksik
etkileri MTT testi ile belirlenmiştir ve en yüksek sitotoksik etki MCF-7 hücrelerinde görülmüştür.
Nanoparçacıkların hücreler üzerinde yarattığı apoptotik etkileri Hücre Akış Sitometresi kullanılarak
değerlendirilmiştir. Apoptotik ölümün ise nanoparçacıklarla 48 saat muamele edilen MCF-7 kanser
hücrelerinde, %93,48’e ulaştığı görülmüştür. Ag nanoparçacıklarının kanser hücrelerine karşı seçici
toksisite ve apoptotik etki göstermesi in vivo kanser tedavisi için umut vaat edici bir sonuçtur ve bununla
birlikte Ag nanoparçacıklarının antioksidan ve antibakteriyel etkinliğinin varlığının da gösterilmesi ile
sentezlenen Ag nanoparçacıklarının potansiyel bir antikanser, antioksidan ve antibakteriyel ajanı olarak
kullanım alanı bulacağı düşünülmektedir.
Nanoparticles are the materials carrying different physicochemical properties and biological activities such as unique chemical reactivity together with different content, size and shapes, and antimicrobial, antiinflammation, antiviral, antioxidant, wound healing, and anticancer activities. Properties of silver (Ag) nanoparticles such as high antimicrobial activity, being a multi-spectrum antibiotic, and low toxicity even at high concentrations make Ag usable in many fields of study. Green synthesis among the nanoparticle synthesis methods, is the frequently used method as an environmentally friendly approach due to its low toxicity together with its ease and low-cost properties. Plant extracts used for this purpose which are already present in nature and do not threaten the body defense mechanism at high extent, are of great interest especially in the field of biotechnology and medicine as materials used for the preparation of multifunctional nanoplatforms with biological origin. Thus, the water extract of the Cotinus coggygria Scop. (Smoke Tree) plant was used in the synthesis of Ag nanoparticles. Nanoparticles with ultra-small sizes and homogenous size distributions and high stability in water could be synthesized by optimizing the reaction parameters in the synthesis. For their characterization, UV/Vis Spectrophotometer, Electron Microscope (STEM), Energy Dispersive X-ray Spectroscopy (EDX), X-ray Diffraction Spectroscopy and Zeta Potential instruments were used. Three kind of breast cell lines (MCF-7, MDA-MB-231 and CRL 4010) were used for in vitro studies of nanoparticles synthesized, the cytotoxic effects of nanoparticles on both cell types were determined by MTT test and the most cytotoxic effect of nanoparticles were observed on MCF-7 cells. The apoptotic effects of nanoparticles on cells were evaluated using Cell Flow Cytometry. It was observed that apoptosis reached %93,48 for MCF-7 cells after treating cells with nanoparticles for 48 hours. The selective toxicity and apoptotic effect of Ag nanoparticles against cancer cells is a promising result for in vivo cancer treatment, and it is thought that Ag nanoparticles synthesized will find use as a potential anticancer, antioxidant and antibacterial agent with the demonstration of the antioxidant and antibacterial activity of Ag nanoparticles.
