dc.description.abstractİnfertil çiftlerde, yardımcı üreme tekniklerinin olumsuz sonuçlarının bir bölümünden ejakülattaki apopitotik spermlerin sorumlu olabileceği düşünülür. İleri düzey sperm hazırlama prosedürlerine rağmen her sperm dölleme sağlamamaktadır. Hyaluronik asit ise oosit etrafındaki kumulus hücrelerinde bulunan bir komponent olup spermin oosite bağlanmasına yardımcı olur. Hyaluronik asite penetre olan sperm sayısının apopitozis ile ters orantılı olacağı düşüncesinden yola çıkarak swim-up medyumuna hyaluronik asit ilavesi ile daha sağlıklı sperm eldesi amaçlanmıştır. 20 hastadan alınan ejakulat örneğine rutin semen analizi yapılmıştır. Her örnek 2 gruba ayrılarak swim-up yöntemi ile hazırlanmıştır. 1. grubun swim-up medyumuna %0,5 Human serum albumin (HSA) 2. gruba ise %10 hyaluronik asit (HA) ilave edilmiştir. Yüzen +3 ve +4 hücre konsantrasyonu belirlenmiş ve annexin V kiti ile boyanıp apopitotik sperm oranı saptanmıştır. Elde edilen +3 ve +4 hücre konsantrasyonu HA içeren grupta HSA içeren gruba göre daha yüksek bulunmuştur. Apopitozis oranı bakımından karşılaştırıldığında HSA içeren grupta; HA içeren gruba göre anlamlı derecede yüksek bulunurken HA içeren grupta en düşük çıkmıştır. Sonuçlarımız, sperm hazırlama medyumuna eklenen hyaluronik asitin sperm parametrelerini olumlu etkileyeceği yönündedir. Bu sayede daha sağlıklı sperm eldesi söz konusudur.en_US
dc.description.abstractDuring assisted reproduction manipulations it is assumed that apoptotic sperms of ejaculate are responsible for failures. Although advanced techniques of sperm preparation harvested every sperm can not be fertile. Hyaluronic acid exists at the cumulus sorrounding oocyte and helps sperm to bind oocyte. Considering that the sperms binding hyaluronic acid shows negative proportion of apoptosis, by adding Hyaluronic acid to swim-up media we searched if a more healthier sperm can be obtained. Routine seminal plasma analysis was done to 20 patients samples. Each sample was divided as two groups; to the swim up medium in group 1; 0,5% HSA in group 2; 10% HA was added. The concentration of +3 and +4 motile sperms were evaluated and with annexin V stain the apopitotic sperm rate was calculated. When +3 and +4 motile sperm rate is compared HA containing group had higher scores. When comparasion is done according to apopitosis HSA containing group had the highest rate with a significant difference to HA containing group, the lowest rate was seen in HA contining group. Out results indicate that by adding HA to sperm processing medium desired sperm parameters are effected positively. By this addition a more healthier sperm population can be harvested.en_US
