Trans-9 18:1 Oktadesenoik Asit İzomerinin İnsülin Direnci Üzerine Etkileri

Özet
Amaç: Trans yağ asitleri hücre membran fonksiyonlarını etkileyebilmektedir. Bu nedenle çalışmamızda Trans-9 18:1 oktadesenoik asit izomerinin insülin direnci üzerine etkilerini araştırmayı amaçladık. Gereç ve Yöntem: Bu çalışmada 40 adet erkek rat kullanıldı. Ratlar dört gruba ayrıldı. 1. Grup yağsız diyetle, 2. Grup oleik asit, 3. Grup margarin ve 4. Grup Trans-9 18:1 oktadesenoik asit izomeri ile 20 gün boyunca beslendi. Beslenme periyodunun ardından ratların yaşamları sonlandırıldı ve kan örnekleri toplandı. Glukoz ve fruktozamin düzeyleri kolorimetrik metodla, adiponektin ve resistin düzeyleri de ELISA metoduyla tesbit edildi. Bulgular: Trans-9 18:1 ile beslenen grupta fruktozamin ve glukoz düzeylerini oleik asit grubuna göre anlamlı şekilde yüksek bulduk. Margarin grubu ile karşılaştırıldığında; oleik asit grubunda fruktozamin, glukoz ve adiponektin seviyeleri anlamlı şekilde düşük, resistin seviyesi anlamlı şekilde yüksek bulunmuştur. Trans yağ asidi grubunda margarin grubuna göre, adiponektin seviyesi anlamlı şekilde düşük, resistin seviyesi ise anlamlı şekilde yüksek çıkmıştır. Sonuç: Tüketilen trans yağ asitlerinin miktarı toplum sağlığı açısından büyük önem taşıdığından diyetteki miktarı mümkün olduğu kadar sınırlandırılmalıdır. Trans yağ asidinin insülin duyarlılığı üzerindeki etkilerini göstermek için kapsamlı çalışmalar yapılması gerekmektedir.
Objective: Since trans fatty acids (TFA) might interfere with cell membrane functions. Therefore, the aim of our study was to investigate the effect of trans 9:18 1 octadecenoic acid isomer on insülin resistance. Materials and Methods: Forty male rats were used in the study. Rats were divided into four groups: group l were fed oil-free diet (control), group 2 were fed oleic acid, group 3 were fed margarine and group 4 were fed trans-9 18:1 octadecenoic acid isomers for 20 days. At the end of the nutritional period, rats were sacrificed and examples of blood was collected. Glucose and fructosamine levels was determinated by colorimetric method, as well as plasma adiponectin and resistin levels by ELISA method. Results: Fructosamine and glucose levels were significantly high in Trans-9 18:1 fed rats compared with oleic acid group. Compared with margarine fed rats, adiponectin, glucose and fructosamine levels were low and resistin level was high in oleic acid group. In TFA group, adiponectin level was significantly low and resistin level was significantly high compared with margarine group. Conclusions: The amount in the diet is very important in terms of public health of the consumed amount of trans fatty acids should be limited as much as possible. Large controlled trials have been required to demonstrate adverse effects of TFA on insülin sensitivity.
