dc.description.abstractSperm motilitesi, erkek infertilitesinde değerlendirilen ilk parametrelerden biri olduğundan in vitro fertilizasyon tedavilerinde motil sperm elde etmek önem arz etmektedir. Bu nedenle farklı yüzdürme metotları birçok çalışmanın konusu olmuş, en ideal yöntemin swim-up olduğu düşünülmüştür. Trombositten Zengin Plazma (TZP), içerdiği büyüme faktörleri nedeniyle ortopedi, plastik cerrahi, kadın doğum gibi farklı alanlarda kullanım bulmuş, fakat üreme ve sperm motilitesi üzerine bugüne kadar yapılmış yalnızca bir çalışma bulunmaktadır. Çalışmamıza semen için 29, kan için 10 gönüllü katılmıştır. Alınan intravenöz kanlardan TZP elde edilmiş, deney protokolünde kullanmak için dondurularak saklanmıştır. 29 gönüllünün rutin semen analizleri yapıldıktan sonra her örnek iki eşit hacimde ayrılmıştır. Eşit hacimde ayrılan bu örnekler raw (ham) hali ve TZP ile muamele edilecek grup olarak iki ayrı şekilde sınıflandırılmıştır. TZP ile muamele edilen semen örneklerinin ve ham grubunun her 15-30 ve 45 dakikada konsantrasyon ve motilite analizleri yapılmıştır. Bulgularımızda elde edilen +4 ve +3 sperm konsantrasyonu ham grubuna göre anlamlı derecede yüksek bulunmuştur (p<.0001). Bu anlamlılığın özellikle ilk 15 dk için kritik olduğu görülmüştür. Sonuçlarımıza göre, TZP'nin sperm motilitesi üzerine anlamlı oranda etkisi bulunmaktadır. TZP, sperm motilitesi düşük IUI ve ICSI için sperm hazırlamada alternatif bir yıkama materyali olabilir.en_US
dc.description.abstractBecause sperm motility is the first parameter evaluated in male infertility, obtaining motile sperm for the in vitro fertilization treatment is considered important. Therefore, different methods of collection have been the subject of many studies and it has been thought that the most ideal method is swim-up. Platelet Rich Plasma has been used in different fields such as orthopaedics, plastic surgery, and gynaecology due to the growth factors it contains, but there is only one study till now on reproductive and sperm motility. For semen 29 volunteers and for blood 10 volunteers participated in our study. Platelet Rich Plasma (PRP) was obtained from the intravenous blood taken and it was frozen for use in the experimental protocol. After routine semen analyses of 29 volunteers were performed, every sample was divided into two equal volumes. These equally divided samples are classified as different groups. One of them is raw form and the other one is the group which will be treated with PRP. Concentration and motility analysis of PRP-treated semen samples and the raw group were performed at each 15th-30th and 45th minutes. Our results showed that the +4 and +3 sperm concentration were significantly higher than the raw group (p<.0001). This significance was found to be critical especially for the first 15 minutes. According to our results, PRP has a significant effect on sperm motility. PRP may be an alternative washing material for sperm preparation to be used in IUI and ICSI which need low sperm motility.en_US
