| dc.description.abstract | Meme kanseri, kadınlarda en çok ölüme neden olan kanser türüdür. Meme kanserinde tümör gelişimine neden olan faktörler iyi anlaşılamamaktadır. Üçlü negatif meme kanserinin tümör agresifliği metastatik ve invaziv özellikleri diğer meme kanseri türlerine göre oldukça kötü prognozludur ve tedavi seçenekleri sınırlıdır. Dolayısıyla, doğal flavonoidlerin ve sentetik türevlerinin antikanser aktivitelerinin araştırılması çok önemlidir. Toll benzeri reseptörler (TLR), patojenik yapılara yanıt verme yetenekleriyle yaygın olarak tanınan korunmuş bir reseptör ailesidir. Meme kanseri ve çevreleyen dokularında TLR genlerinin ifadesi değişen derecelerde yukarı veya aşağı düzenlenir. Meme kanserinde, TLR2 ve TLR4 ifadesi oldukça yüksek olması nedeniyle hücrenin apoptoza girmesine engel olmaktadır. Dolayısıyla bu durum yüksek mortalite ile ilişkilendirilmektedir. Robinin TLR yolağının önemli üyelerinden TLR2 ve TLR4’ün antagonistidir. Ancak, meme kanserinde TLR, apoptoz ve ER stresi ilişkili yolaklara etkisi henüz bilinmemektedir. Bu çalışma da; MDA-MB-231 meme kanseri hücrelerinde Robinin’in antikanser etkisi araştırılmıştır. Hücre proliferasyonuna etkisi CCK-8 sitotoksisite deneyi ile değerlendirilmiştir ve 48. saatte IC50 değeri 16,67 µM olarak belirlenmiştir. Daha sonra Robinin’in meme kanseri hücrelerinde TLR2, TLR4, apoptoz (BAX, BCL2, CASP3, CASP7, CASP8, CASP9, CYCS, FADD, FAS ve P53) ve ER stres (ATF4, ATF6, CALR, CHOP, EIF2A, GRP78, HSP47, IRE1, PERK ve XBP1) sinyal yolaklarında bulunan gen ifadeleri üzerine etkinliği qRT-PCR ile tespit edilmiştir. BAX, CASP3, CASP7, CASP8, CYCS, FADD, ATF4, CALR, CHOP, EIF2A, GRP78, IRE1 ve XBP1’de anlamlı artış gözlenirken BCL2’nin baskılandığı tespit edilmiştir. Ayrıca mRNA düzeyinde ifadesi anlamlı değişen bazı genlerin protein düzeyleri ELISA ve western blot testi ile değerlendirilmiştir. BAX, CASP3, CASP7, CASP8, CYCS ve P53 protein seviyesinde anlamlı artış olduğu görülmüştür. Robinin’in etkisinin in vivo çalışmalar ile desteklenerek moleküler düzeyde belirlenmesi, meme kanserinin adjuvan tedavisinde yeni bir ajan olabileceği düşünülmektedir. | |
| dc.description.abstract | Breast cancer is the most common type of cancer that causes death. The factors that cause tumors in breast cancers are not well understood. The tumor aggressiveness, metastatic and invasive features of triple negative breast cancer have a very poor prognosis compared to other types of breast cancer and treatment options are often limited. Therefore, it is very important to investigate the anticancer properties of natural flavonoids and their synthetics. Toll-like receptors (TLR) are a type of receptor that is widely defined by its ability to respond to pathogenic structures. In breast cancer and its surrounding tissues, the expression of TLR genes are up- or downregulated to varying degrees. In breast cancer, TLR2 and TLR4 expression is quite high, preventing the cell from entering apoptosis, which is thereby associated with high mortality. Robinin is an antagonist of TLR2 and TLR4, which are known as important members of the TLR pathway. However, effects of Robinin on TLR, apoptosis and ER stress-related pathways in breast cancer are not yet known. In this study, the anticancer effects of Robinin were investigated in MDA-MB-231 breast cancer cells. Its effect on cell proliferation was evaluated by CCK-8 cytotoxicity assay and the IC50 value was determined as 16.67 µM at 48 hours. The effects of Robinin on gene expression of TLR2, TLR4, apoptosis (BAX, BCL2, CASP3, CASP7, CASP8, CASP9, CYCS, FADD, FAS and P53) and ER stress (ATF4, ATF6, CALR, CHOP, EIF2A, GRP78, HSP47, IRE1, PERK and XBP1) signaling pathways were evaluated using qRT-PCR. Significant increases were observed in BAX, CASP3, CASP7, CASP8 CYCS, FADD, ATF4, CALR, CHOP, EIF2A, GRP78, IRE1 and XBP1 expression levels. However, BCL2 was downregulated. In addition, ELISA and western blot tests were performed on genes that were significant changed at the mRNA level. A significant increase was observed in the protein levels of BAX, CASP3, CASP7, CASP8, CYCS and P53. Determination of the effect of robinin at the molecular level supported by further in vivo studies is thought to be a potential novel agent in the adjuvant treatment of breast cancer. | |
