Öğe 3?,4?-Dihydroxyflavonol (DiOHF) prevents DNA damage, lipid peroxidation and inflammation in ovarian ischaemia-reperfusion injury of rats(Taylor & Francis Inc, 2022) Agartan, Elif Sena; Mogulkoc, Rasim; Baltaci, Abdulkerim Kasim; Menevse, Esma; Dasdelen, Dervis; Avunduk, Mustafa CihatThis study aimed to determine the effect of 3',4'-Dihydroxyflavonol (DiOHF) on lipid peroxidation, DNA damage and inflammation in ovarian ischaemia (I)-reperfusion (R) injury. This study was performed on 44 Wistar-albino female rats. Groups were designed as Control; Sham; I/R (the left ovary was ligated for 2 h and then reperfused for 2 h); I/R + DiOHF (after 2 h ischaemia and 2 h reperfusion, 30 mg/kg of DiOHF was given intraperitoneally and reperfusion was allowed for 2 h more); I + DiOHF + R (after 2 h I, 30 mg/kg of DiOHF was given at the beginning of 2 h reperfusion); DiOHF + I/R (2 h after DiOHF administration, the left ovary was ligated for 2 h and then reperfused for 2 h). Blood and ovarian tissue samples were analysed for GSH, MDA, 8-OHdG, SOD, and IL-6. Ovarian tissue was examined histopathologically. Ovarian I/R has led to inflammation and oxidative damage. However, DiOHF activated the antioxidant system and prevented DNA damage induced by I/R in ovarian tissue. Vascularisation, oedema, and inflammation also occurred in ovarian tissue in I/R group. The results of this study indicated that I/R led to disturbance of the oxidant/antioxidant system balance and increased DNA damage; however, DiOHF supplementation prevented DNA damage, lipid peroxidation and inflammation by increasing the antioxidant system in ovarian I/R injury in rats. However, in potential I/R situations, DiOHF application appears to be beneficial in reducing inflammation, oxidant injury, and DNA damage, and in activating the antioxidant system. IMPACT STATEMENT What is already known on this subject? Ischaemia/reperfusion (I/R) injuries lead to damage in cells or tissues due to insufficient blood flow. What do the results of this study add? Increased DNA injury and inflammatory response (IL-6) and structural impairment were treated by administration of intraperitoneal (DiOHF) which strongly stimulated the antioxidant system, inhibited antioxidant activities, prevented DNA damage and inflammation process. What are the implications of these findings for clinical practice and/or further research? This study's strength is that it is the first research demonstrates the prevention of DNA damage in ovarian I/R by DiOHF supplementation. This flavonoid (DiOHF) may be used for treatment in different ovarian ischaemia/reperfusion.Öğe EFFECTS OF CURCUMIN TREATMENT ON CELL ENERGY STATUS, LEVELS OF MITOCHONDRIAL ENZYMES, AND GENE EXPRESSION OF GLUCOSE-RELATED MECHANISM IN PANCREATIC CANCER CELL LINES(Dokuz Eylul Univ Inst Health Sciences, 2023) Korucu, Emine Nedime; Menevse, Esma; Kaya, Dudu Erkoc; Gokturk, Fatma; Arikoglu, HilalPurpose: Curcumin is an active component of turmeric, has antitumor, immunomodulatory, anti-inflammatory effects. This study aimed to investigate the effects of the administration of curcumin on the energy metabolism, the abnormal redox defense mechanism profile, the malignant transformation indicator of Panc-1 and BxPC-3 pancreatic cancer cells.Material and Methods: BxPC-3 and Panc-1 cells were treated with various concentrations of curcumin (20 mu M, 40 mu M, 50 mu M, 60 mu M, 80 mu M, 100 mu M, and 125 mu M ) for 24 h. Cell lysate Adenosine triphosphate (ATP), Adenosine diphosphate (ADP), Adenosine monophosphate (AMP), Manganese superoxide dismutase (MnSOD), and cytochrome p450 reductase (CPR) concentrations were analyzed with HPLC and ELISA methods. Genes expression of Lactate dehydrogenase (LDH), mitochondrially encoded ATP synthase membrane subunit 6 (MTATP6), Glucose transporter 1 (GLUT1), and cytochrome p450 were analyzed by real-time quantitative PCR.Results: IC50 values for 24 hours were found as 47.26 mu M in BxPC-3 and 45.84 mu M in Panc-1 cells. Treatment with curcumin inhibited oxidative stress by increasing MnSOD enzyme levels. ATP levels did not change in BxPC-3 cells, but they increased in Panc-1 supplemented with curcumin. Gene expression of GLUT-1 was significantly down regulated when using at 45 mu M concentration of curcumin, which also affected glucose consumption in both cells. Conclusion: Curcumin showed anti-proliferative and antioxidant effects.Öğe Evaluation of sialic acid, malondialdehyde and glutathione levels in infertile male(Elsevier Espana Slu, 2022) Menevse, Esma; Dursunoglu, Duygu; Cetin, Nihal; Korucu, Emine Nedime; Erbayram, Fatma ZehraIntroduction and objectives: Around 15% of productive couples in the world are infertile. Recent years, biochemical mechanisms leads to male infertility are started to research. Redox regulation and oxidative stress (OS) show importance in the pathogenesis of infertility in male. Malondialdehyde (MDA) and Glutathione (GSH) are biochemical indicatives of sperm damage by reactive oxygen species (ROS). In addition, sperm are coated with a thick glycocalyx rich in sialic acids. It is aimed to determine and evaluate the differences between normozoosper-mic and oligozoospermic individuals according to sialic acid, MDA and GSH concentrations and correlations between spermyogram and these parameters. Material and methods: This study was carried out on seminal plasma of individuals who admit-ted to Selcuk University Faculty of Medicine IVF Unit Andrology Laboratory. The groups were divided into two as normozoospermics (n = 30, sperm concentration >= 15 million/mL), and oligozoospermics (n = 30, sperm concentration < 15 million/mL). Spermyogram were evaluated regarding WHO (2010) Kruger criteria. GSH, MDA and sialic acid concentrations were analyzed in seminal plasma. Diagnostic performance of sialic acid has been determined with ROC curve analysis. Results: Sialic acid levels were significantly lower in Normozoospermic than Oligozoosper-mic individuals (p < 0.0001), MDA and GSH levels were not differ significantly in both groups (p > 0.05). Sialic acid correlated significantly with most of the spermyogram findings. When diagnostic performance of sialic acid was evaluated, the cut off value of sialic acid found as 4.175 nmol/mL by ROC curve. Conclusion: High seminal plasma sialic acid levels may be used as a biomarker and sialic acid is important determinant in oligozoospermia. All rights reserved.Öğe Evaluation of the effects of ankaferd blood stopper on rabbits with paranchyme damage: an experimental study(Baycinar Medical Publ-Baycinar Tibbi Yayincilik, 2013) Metin, Bayram; Altinok, Tamer; Menevse, Esma; Esen, HasanBackground: In this study, we aimed to evaluate the effects of Ankaferd Blood Stopper (ABS), a herbal extract used as a hemostatic agent in traditional Turkish medicine, on hemostasis, wound healing, air leakage and the efficiency over pleural adhesions in rabbits with pulmonary parenchymal damage. Methods: Sixteen New Zelland Albino type male and female rabbits with a mean weight of 2500 g were used. The rabbits were divided into two groups as study group and control group. One rabbit in each group died during anesthesia. We performed wedge resections in equal size to the left lower lobes of all rabbits. No additional procedure was performed on the control group, while the study group received 5 puffs (similar to 1 cc) of ABS to the resection area. Tube thoracostomy, which was performed in both groups, was terminated postoperatively at the sixth hours after drainage and air leakage follow-up. The rabbits were sacrificed with high dose anesthetic at the postoperative eight-day. Pleural inflammation and fibrosis in the hemithorax were examined macroscopically and microscopically. Biochemical measurements of tissue malondialdehyde (MDA) and hydroxyprolin (HPR) levels were done. Results: There was a statistically significant difference in postoperative air leakage and the amount of drainage between the groups (amount of hemorrhage p<0.05, air leakage p<0.001). A higher rate of macroscopic and microscopic fibrosis was observed in the study group. When both groups are compared according to tissue MDA levels, decreased MDA level was seen in the group which ABS was administered, where the difference was statistically non-significant. Conclusion: Ankaferd Blood Stopper reduces postoperative bleeding and prolonged air leakages without any toxic effects and has an effective role at pleurodesis, increasing the pleural fibrosis level.Öğe Investigation of the effects of the toll-like receptor 4 pathway on immune checkpoint vista in pancreatic cancer(Springer, 2022) Topcu, Kubra Sena Bas; Korucu, Emine Nedime; Menevse, Esma; Kocak, Nadir; Duran, TugcePancreatic ductal adenocarcinoma (PDAC) is one of the most common malignant tumors of the pancreas. Preclinical studies show that it evades the immune system with immune checkpoints and promotes tumor development. V-domain Ig suppressor of T cell activation (VISTA) is a new immune-check point from the B7 family and is highly expressed in cancer cells. Overexpression of toll like receptor 4 (TLR4) in pancreatic adenocarcinoma is associated with induced tumorigenesis, tumor growth, resistancy to chemotherapy. Naloxone is an opioid and inhibits TLR4-ligand association. In this study, we investigated the relation of TLR4 and downstream pathways with immune-check point VISTA in pancreatic cancer proliferation. We initially collected pancreatic cancer-related datasets using the GEPIA2 and UALCAN databases. Based on this data obtained the effect of various concentrations and incubation times of naloxone were used on PANC-1 cells proliferation. A combination of naloxone and VISTA-siRNA were applied, and the effect of both naloxone and combined treatment on TLR4, Interleukin 1 receptor associated kinase 4 (IRAK4) and VISTA gene expression were analyzed in pancreatic cancer cells. As a result of analysis with Quantitative Real-Time Polymerase Chain Reaction (qRT-PCR), gene expression levels of TLR4, IRAK4 and VISTA were significantly suppressed and cell proliferation was significantly reduced. We found that administration of naloxone and VISTA-siRNA in combination with PDAC cells suppressed signaling. Therefore, we considered that the relationship between VISTA and TLR4 signaling pathways and the other possible associated signal molecules may be an important marker in determining the response of immune checkpoint inhibitors in cancer treatment.Öğe THE LEVELS OF ADENOSINE TRIPHOSPHATE SYNTHASE 6, MANGANESE SUPEROXIDE DISMUTASE, NADPH-CYTOCHROME P450 REDUCTASE, LACTATE DEHYDROGENASE IN SEMINAL PLASMA OF OLIGOZOOSPERMIA AND NORMOZOOSPERMIA MEN(Nobel Ilac, 2023) Menevse, Esma; Dursunoglu, Duygu; Korucu, Emine Nedime; Cetin, Nihal; Erbayram, Fatma ZehraObjective: In recent years, energy production and enzymes in mitochondrial pathways are an important in male fertilization.Material and Method: For this purpose, we tried to determine the relationship between normozoospermia and oligozoospermia individuals according to levels of some mitochondrial enzymes. The seminal plasma levels of mitochondrial encoded adenosine triphosphate synthase 6 (mtATP6), manganese superoxide dismutase (Mn-SOD), NADPH-cytochrome P450 reductase (CPR), lactate dehydrogenase (LDH) were analyzed and assessed their correlations between data of spermogram. The study was composed of two groups consisting of normozoospermic and oligozoospermic volunteers.Results: mtATP6 levels were significantly higher in normozoospermia (0.97 & PLUSMN;0.50 ng/mg protein, p=0.008) than oligozoospermia (0.64 & PLUSMN;0.32 ng/mg protein). LDH levels were higher in normozoospermia (0.25 & PLUSMN;0.074 U/ mg protein, p=0.007) than oligozoospermia (0.199 & PLUSMN;0.049 ng/mg protein). The groups did not differ in terms of MnSOD and CPR levels. mtATP6 levels were significantly correlated with sperm concentration, total number and total motility, immotility, total progressive motile sperm count (TPMSC) and long head anomaly. LDH levels were related with long head and short tail anomaly.Conclusion: We observed some of the spermatogenetic anomalies are caused by defects in energy regulation. We did not find any evidence on MnSOD and CPR activity regarded as determinative parameters in response to increased oxidative stress in oligozoospermia.
