Öğe Assessment of Structure, Function, and Rhythm of the Heart with Echocardiography and Electrocardiography in Adolescent Swimmers(Springer, 2021) Akkus, Abdullah; Belviranli, Muaz; Sap, Fatih; Okudan, NilselThe aim of this study was to evaluate the cardiac parameters by using electrocardiography and echocardiography in adolescent swimmers. Twenty-two adolescent swimmers and 22 gender- and age-matched sedentary controls admitted to our center between November 2018 and May 2019 were included in this study. In addition to demographical characteristics, participants were assessed via a 12-lead electrocardiography and two-dimensional echocardiography for cardiac function. On the echocardiography, end-systolic and end-diastolic interventricular septum, end-systolic and end-diastolic left ventricular posterior wall thicknesses, left atrial width, Tricuspid E, left ventricular mass and left ventricular mass index were higher in the swimmers when compared to the sedentary controls (P < 0.05). On the electrocardiography, Tp-e duration which reflects ventricular transmural repolarization, and Tp-e/QT and Tp-e/corrected QT ratios were higher in the swimmers than the sedentary controls (P < 0.05). In conclusion, swimming exercise in children leads to concentric thickening of left ventricle and induces an increase in Tp-e duration, and Tp-e/QT and Tp-e/corrected QT ratios, which are the novel markers for risk of ventricular arrhythmias.Öğe Curcumin improves spatial memory and decreases oxidative damage in aged female rats(Springer, 2013) Belviranli, Muaz; Okudan, Nilsel; Atalik, Kismet Esra Nurullahoglu; Oz, MehmetAging induced cognitive impairment has been well documented for many years and several antioxidant strategies have been developed against this impairment. Curcumin is the active component of curcuma longa and has shown antioxidant, antiinflamatory and neuroprotective properties. We hypothesized that curcumin would have an influence on cognitive functions in aged female rats. The purpose of the present study was to investigate the effects of curcumin supplementation on cognitive impairment evaluated by Morris water maze (MWM) as well as the oxidative stress induced by aging in female rats. Rats were randomly divided into either control or curcumin-supplemented groups. Curcumin or vehicle (corn oil) were given once daily for a period of 12 days, beginning 7 days prior to and 5 days during the behavioral tests. Behavioral assessment was performed in MWM. At the end of the behavioral test, blood samples and brain tissues were taken for the analysis of malondialdeyde (MDA), protein carbonyl and glutathione levels. During the training session, curcumin supplementation decreased latency to reach to the platform and the total distance traveled. During the probe trial, curcumin supplementation increased the number of platform crossings. In addition to the behavioral testing, biochemical results showed that MDA levels decreased in brain tissue by curcumin supplementation. It may be concluded that, curcumin supplementation improves cognitive functions by decreasing the lipid peroxidation in brain tissue of aged female rats.Öğe Dört haftalık koenzim Q10 desteğinin sedanter genç erkeklerde egzersizle oluşan kas hasarı üzerine etkileri(2012) Torlak, Savaş; Okudan, Nilsel; Gökbel, Hakkı; Belviranlı, Muaz; Kıyıcı, AyselAmaç: Bu çalışmanın amacı, sedanter genç erkeklerde ekzantrik egzersizin neden olduğu kas hasarında koenzim Q10 (CoQ10) takviyesinin etkilerini incelemekti. Yöntem: Çalışmaya 21 sağlıklı sedanter genç erkek katıldı. Tüm katılımcıların boyları, vücut ağırlıkları ve vücut yağ yüzdeleri ölçüldü ve istirahat kanları alındı. Dört hafta süreyle 11 katılımcıya oral yolla günde tek doz 200 mg CoQ10, 10 katılımcıya plasebo verildikten sonra ölçümler ve kan alımı tekrarlandı. Daha sonra bütün katılımcılara izokinetik egzersiz dinamometresinde ekzantrik egzersiz protokolü uygulandı ve egzersiz sonrası 24. ve 48. saatte kan örnekleri alındı. Alınan kan örneklerinde plazma koenzim Q10 seviyesi, serum kreatin kinaz aktivitesi ve serum miyoglobin seviyesi tayin edildi. Bulgular: Plazma CoQ10 seviyesi CoQ10 takviyesi yapılan grupta plasebo grubuna göre daha yüksekti. Kreatin kinaz aktivitesi ve miyoglobin seviyesi egzersiz sonrası 24. ve 48. saatte her iki grupta da arttı, ama her iki grup arasında anlamlı fark yoktu. Sonuç: Dört haftalık koenzim Q10 takviyesi kan koenzim Q10 seviyesini artırırken egzersizle oluşan kas hasarını etkilemez.Öğe The effect of isotretinoin therapy on oxidative damage in rats(Wiley, 2020) Daye, Munise; Belviranli, Muaz; Okudan, Nilsel; Mevlitoglu, Inci; Oz, MehmetIsotretinoin is prescribed in many dermatologic disorders, but mostly in acne. There is limited research about oxidative stress induced by isotretinoin and its effects on the liver tissue, muscle tissue, and blood. In this study, oxidative damage of isotretinoin on the liver, muscles, and blood in rats at the therapeutic dosage for humans, is evaluated. Thirty, 2-months-old Wistar albino rats were randomly divided into four groups. Isotretinoin was administered at the human equivalent low dose of 7.5 mg/kg by gavage. Blood, liver, and skeletal muscle samples were taken from the animals under anesthesia. Oxidative stress and antioxidant defense markers such as Malondialdehyde (MDA), Protein carbonyl (PC), 8-OHDG (8-hydroxy-deoxyguanosine), SOD (Superoxide dismutase), GSH(Glutathione), GPX (glutathione peroxidase), NO (Nitric Oxide) levels, Aspartate aminotransferase (AST), Alanine aminotransferase (ALT), and creatine kinase (CK) levels were measured. There were significant differences between the ALT values of the control group and the third month of isotretinoin treatment group. Oxidative stress markers such as 8-OHDG, PC, GSH, GPX, and NO values significantly differed in month 3. SOD was significantly lower in the treatment groups compared to the control group. Our study supports that the levels of oxidative markers are increasing with the isotretinoin treatment so this may flare acne. GPX levels increased at the muscle tissue level, and may be responsible for the myopathy that is seen in acne patients. Beginning on day 7 after STZ injection, the rats were administered GSE (100 mg kg(-1) day(-1)) in drinking water for 6 weeks. At the end of week 6, rats were sacrificed by cardiac puncture. Plasma nitric oxide (NO) levels and xanthine oxidase (XO), adenosine deaminase (ADA), and glutathione peroxidase (GPx) activities were analyzed. Results: Both XO and ADA activities increased and NO levels decreased in diabetic rats (P < 0.05). GSE supplementation normalized all of these changes. Antioxidant enzyme activities decreased in diabetic rats compared to the controls (P < 0.05). GSE supplementation increased antioxidant enzyme activities in both diabetic and healthy rats (P < 0.05). The following tests are generally used in pulmonary diseases: Stair Climbing Test: It is the easiest and the most economic clinical exercise test. It is used for the assessment of functional capacity in patients with chronic obstructive pulmonary disease (COPD) and post-operative complication risk in thoracic surgery. Six-Minute Walk Test: This is the most common exercise test for evaluating pulmonary diseases. The advantage of the test is that patients can complete the test by increasing their speed if respiratory insufficiency occur during the test. Shuttle Test: Two different types of the tests are incremental shuttle and endurance shuttle walking tests. Incremental shuttle walking test is used for the determination the peak oxygen consumption. During the endurance shuttle walking the speed is held constant. This test is suitable for the assessment of the pulmonary rehabilitation programs. Exercise-Induced Bronchospasm Test: Generally a 6 to 8-minutes trenuous exercise, which increases the FEV1 approximately 20fold, is required to estimate exercise-induced bronchospasm. Cardiopulmonary Exercise Testing (CPET): CPET is accepted as gold standard in the evaluation of exercise intolerance. It is used in the preparation of rehabilitation programs and exercise prescriptions and evaluation of the pre-andpost-surgical condition. Systemic responses to the exercises are investigated best by incremental exercise tests. There fore, two equipments; the treadmill and the cycle ergometer, can be used. Values of VO2, VCO2, oxygen saturation, heart rate and systemic blood pressure, and ECG can be followed during CPET. During the test it is important to evaluate patient's perceptions about the test, dyspnea, chest pain and fatigue and to determine the limit of the workload when patients can not continue to the test.Öğe Psoriasisli Hastalarda Serum Nöroinflamatuvar Belirteç Seviyeleri ve Geniş Çaplı Periferik Sinir Lifi Fonksiyonlarının Değerlendirilmesi(2016) Balevi, Ali; Özdemir, Mustafa; Güney, Figen; Tol, Hüseyin; Okudan, Nilsel; Belviranlı, Muaz; Şahin, Kemal TahirAmaç: Günümüzde tümör nekroz faktör-alfa (TNF-?) inhibitörleri, özellikle şiddetli psoriasisin tedavisinde tüm dünyada yaygın olarak kullanılmaktadırlar. Ancak psoriasisli hastalarda TNF-? inhibitör tedavisiyle ilişkilendirilmiş multipl sklerozis (MS) ve periferal nöropatiler bildirilmektedir. Psoriasis tedavisinde kullanılan TNF-? inhibitörleri, altta yatan nörolojik bozukluğu ortaya çıkarıp çıkarmadığı bilinmemektedir. Bu çalışmanın amacı; psoriasis ile multipl sklerozis ve periferal nöropatiler arasında bağlantı olup olmadığını araştırmaktır. Yöntemler: Bu çalışmada; 50 psoriasisli hasta ve 25 sağlıklı kontrol çalışmaya alındı. Nörolojik muayene ve iğne elektromiyografi testleri uygulandı. Nöroinflamatuvar süreçte rol alan matriks metalloproteinaz-2 (MMP-2), matriks metalloproteinaz-9 (MMP-9), S100 kalsiyum bağlayıcı protein (S100B) ve nöron spesifik enolaz (NSE) seviyeleri serumda ölçüldü. Bulgular: Sağlıklı kontrol grubu ile karşılaştırıldığında serum MMP-9 seviyeleri psoriasisli hastalarda yüksek tespit edilirken MMP-2 ve NSE seviyeleri psoriasisli hastalarda, sağlıklı kontrollere göre düşük tespit edildi. Serum S100B seviyeleri açısından her 2 grup arasında fark yoktu. Tüm hastalarda ve sağlıklı kontrollerde tüm nörofizyolojik parametreler normal bulundu ve 2 grup arasında fark tespit edilmedi. Sonuç: NSE ve MMP-9, psoriasisli hastalarda MS'in oluşmasında rol oynuyor olabilir. Özellikle ailesel MS öyküsü olan psoriasisli hastalarda, TNF-? inhibitörü ajan başlamadan önce bu belirteçlerin ölçülmesinin faydalı olabileceğini düşünüyoruz.Öğe Serum Neuroinflammatory Markers' Levels and Evaluation of Peripheral Large Nerve Fibre Function in Patients with Psoriasis(Galenos Yayincilik, 2016) Balevi, Ali; Ozdemir, Mustafa; Guney, Figen; Tol, Huseyin; Okudan, Nilsel; Belviranli, Muaz; Sahin, Kemal TahirObjective: Currently, tumor necrosis factor-alpha (TNF-alpha) inhibitors are widely used all over the world, especially in the treatment of severe psoriasis. However, multiple sclerosis (MS) and peripheral neuropathy occurrence associated with TNF-a inhibitors have been reported in the treatment of patients with psoriasis. It is not known whether TNF-a inhibitors reveal underlying neurological disorders in patients with psoriasis. The aim of this study was to investigate whether there is an association between psoriasis and MS or peripheral neuropathy. Methods: In this study, 50 psoriasis patients and 25 healthy controls were enrolled. Neurological examination and needle electromyography test were performed. Serum levels of matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-9 (MMP-9), S-100 calcium-binding protein (S100B) and neuron specific enolase (NSE), which are involved in neuroinflammatory process, were measured. Results: Serum MMP-9 levels were higher in psoriasis patients compared to healthy controls. Serum MMP-2 and NSE levels were decreased in psoriasis patients compared to healthy controls. There was no significant difference in S100B levels between two groups. All neurophysiological parameters in each individual patient and control were found to be within normal limits and there was no difference between groups. Conclusion: NSE and MMP-9 might play role in the occurrence of MS in psoriasis patients. Especially, we are in the opinion that it might be useful to measure the serum levels of these markers before the administration of TNF-a inhibitors in psoriasis patients with family history of MS.Öğe Single and repeated sevoflurane or desflurane exposure does not impair spatial memory performance of young adult mice(Wiley, 2013) Kilicaslan, Alper; Belviranli, Muaz; Okudan, Nilsel; Atalik, Esra NurullahogluVolatile anesthetics are known to disturb the spatial memory in aged rodents, but there is insufficient information on their effects on young adult rodents. The aim of this study was to compare the effects of single and repeated exposure to desflurane and sevoflurane on spatial learning and memory functions in young adult mice. Balb/c mice (2months old) were randomly divided into six equal groups (n=8). The groups with single inhalation were exposed to 3.3% sevoflurane or 7.8% desflurane or vehicle gas for 4h, respectively. The groups with repeated inhalation were exposed to 3.3% sevoflurane or 7.8% desflurane or vehicle gas for 2h a day during 5 consecutive days. Spatial learning and memory were tested in the Morris water maze 24h after exposure. In the learning phase, the parameters associated with finding the hidden platform and swimming speed, and in the memory phase, time spent in the target quadrant and the adjacent quadrants, were assessed and compared between the groups. In the 4-day learning process, there was no significant difference between the groups in terms of mean latency to platform, mean distance traveled and average speed (P>0.05). During the memory-test phase, all mice exhibited spatial memory, but there was no significant difference between the groups in terms of time spent in the target quadrant (P>0.05). Sevoflurane and desflurane anesthesia did not impair acquisition learning and retention memory in young adult mice.Öğe Yaşlı sıçanlarda antrenman ve koenzim Q10 takviyesinin kalp dokusunda oksidatif stres ve antioksidan savunma belirteçleri üzerine etkileri(2015) Belviranlı, Muaz; Okudan, Nilsel; Özdemir, Ayşe Nurten; Öz, MehmetBu çalışmanın amacı, yaşlı sıçanlarda antrenman ve/veya koenzim Q10 (CoQ10) desteğinin kalp dokusunda oksidatif stres ve antioksidan savunma değişiklikleri üzerine etkilerini incelemekti. Gereç ve yöntem: Çalışmada 27 genç (3 aylık) ve 27 yaşlı (20 aylık) Wistar erkek sıçan kullanıldı. Her bir grup; kontrol, antrenman, CoQ10 ve CoQ10antrenman olmak üzere dörder alt gruba ayrıldı. Antrenman gruplarındaki sıçanlara 8 hafta boyunca, haftada 5 gün, günde 1 saat yüzme egzersizi yaptırıldı. CoQ10 desteği alan sıçanlara 8 hafta boyunca günde 300 mg/kg CoQ10 oral olarak verildi. Sıçanlar, anestezi altında kalp dokusu örnekleri alındıktan sonra servikal dislokasyonla sakrifiye edildi. Alınan örneklerde malondialdehit (MDA), 8-hidroksi-2-deoksiguanozin (8-OHdG), glutatyon (GSH) seviyeleri ve süperoksit dismutaz (SOD) ve katalaz (CAT) aktiviteleri ölçüldü. Bulgular: Gruplar arasında MDA, 8-OHdG ve GSH seviyelerinde herhangi bir fark yoktu. SOD ve CAT aktivitesi genç CoQ10 ve CoQ10antrenman gruplarında ve tüm yaşlı gruplarda, genç kontrol ve genç antrenman gruplarından düşüktü. Sonuç: Yaşlanma, CoQ10 takviyesi ve antrenman, kalp dokusunda lipid peroksidasyonu, DNA hasarı ve enzimatik olmayan antioksidan seviyeleri üzerine sınırlı etkiye sahipken, antioksidan enzim aktivitesini azaltırÖğe Yaşlı sıçanlarda kurkumin takviyesinin kalp dokusunun oksidan/antioksidan durumu üzerine etkileri(2012) Belviranlı, Muaz; Okudan, Nilsel; Nurullahoğlu Atalık, Kısmet EsraAmaç: Çalışmanın amacı kurkumin takviyesinin yaşlı dişi sıçanların kalp dokusunda oksidatif stres ve antioksidan savunma üzerine etkilerini araştırmaktı. Yöntem: Çalışmada 20 aylık 20 dişi Wistar sıçan kontrol ve kurkumin grupları olmak üzere rastgele 2 gruba ayrıldı. Kurkumin grubuna 12 gün boyunca günde 300 mg/kg dozunda kurkumin mısır yağı içerisinde çözülerek oral gavaj yoluyla verildi. Son takviyeden 24 saat sonra anestezi altında kalp dokuları alındı ve alınan örneklerde malondialdehit (MDA), protein karbonil (PC) ve glutatyon (GSH) seviyeleri analiz edildi. Bulgular: Kurkumin takviyesi GSH seviyelerini önemli ölçüde artırmıştır (P<0,05). Bununla birlikte, kurkumin takviyesi istatistiksel olarak anlamlı olamamasına rağmen MDA ve PC seviyelerini azaltmıştır (P>0,05). Sonuç: Kurkumin takviyesi yaşlı dişi sıçanlarda kalp dokusunu oksidatif hasara karşı korur ve antioksidan savunma sistemini güçlendirir.
